So "thedude"'s question has got me inspired. I've never been to both though am curious how they shape up against each other as they're both urban metropolises that blend Western and Eastern culture together.
I've been to both places and I would choose Hong Kong over Singapore. Singapore is that place with the world's biggest ferris wheel right? Yeah, I've been on that too. Plus, in addition to that, Hong Kong has the ferry rides and the speed boats that could take you to Macau! (!
He he, I said: "speed"!
You can easily compare them : they both suck.

never been to Singapore but I didn't enjoy HK, Big cities in China are nice and new but a city is a city to me. I like seeing old or rural China.
Singapore is like a sterile shopping center, it has a lot of good things, it's clean, safe etc, and it is easy to get to Malaysia.
Hong Kong has a lot more personality, history and a lot of China's craziness, but in a structured form.
I wanted to visit Singapore. Then I read about NEWater.
I haven't been able to shake off this ugly image I've had of Singapore ever since, so I never went to Singapore.
Hong Kong is great. It is much more livable than Beijing. You don't fear for your life when you want to cross the street. Things in Hong Kong can be pricey, but not overpriced. Bars/Clubs have much better music. Lan Kwai Fong is much classier than Sanlitun.
I don't mind big crowds, other people might not enjoy Hong Kong because of this.

My family has a nice farm with access to clean water. I'd rather live on our farm than drive a Ferrari in Singapore.
I haven't seen a lot of either city, but from my initial impressions, there's a far greater disparity between rich and poor in HK... I didn't see as much of a shanty town in Singapore as the one's I've noted in HK. (could be I wasn't in the right - or wrong - place).
Singapore also has a far greater Indian and Malaysian influence (not surprisingly), while HK is obviously a mix of Chinese and English. This does make for a different atmosphere... Singapore appeared more laid back and relaxed. However, due to issues with Malaysia, there's this little... thing.. in the locals that you don't get in HK... a sort of paranoia.
But, they're both nice places to go to (except in the summer!!!)
